    Effects of Tannin Part of Tibetan Medicine Phyllanthi Fructus on Apoptosis and Cell Cycle of Human Tumor Cells

    Objective To investigate the mechanism of antitumor activity of tannin part of Tibetan medicine Phyllanthi Fructus on human tumor cells in vitro. Methods The human tumor A549 cells and BEL-7402 cells were treated with tannin part of Tibetan medicine Phyllanthi Fructus with 40, 80 and 160 μg·mL-1 for 48 h. They were detected by MTT assay.Morphology changes in cells were examined under the inverted phase contrast microscope. Cell cycle and apoptosis in both cells were determined by flow cytometry. Results MTT assay showed a significant cytotoxicity on A549 cells and BEL-7402 cells in different concentration of tannin part of Tibetan medicine Phyllanthi Fructus for 48 h. Significant growth inhibition and cells shrinkage were observed in cells that received treatment. Significant cells early apoptosis and G2/M phase block were induced by tannin part of Tibetan medicine Phyllanthi Fructus. Conclusion Tannin part of Tibetan medicine Phyllanthi Fructus showed antitumor activity in vitro. Inducing tumor apoptosis and cell cycle block contribute to this activity.

    Effects of Arctigenin Injection on the Cardiovascular System and Respiratory System in Conscious Beagle Dog

    Objective To evaluate the effects of arctigenin injection(NBZGY)on the cardiovascular system and the respiratory system by subcutaneous administration in conscious Beagle dogs that implanted with telemetry transmitters. Methods Eight Beagle dogs with telemetry transmitters(TL11M2-D70-PCTR)were subcutaneously administrated with NBZGY injection (0.05~5 mg·kg-1) and 30% PEG (normal control). All dogs received normal control and NBZGY injection in sequence every other day. The ECG (RR interval, heart rate, T wave, ST segment, QRS interval, PR interval, QT interval), body temperature, blood pressure (systolic blood pressure, diastolic blood pressure, mean blood pressure) and respiratory (tidal volume, respiratory rate) data were continuously monitored for at least 2 h before dosing and 1, 1.5, 2, 3, 4, 5, 6, 12 and 24 h after dosing. Results Compared with the vehicle control group at 1, 2, 24 h after dosing, tidal volume, respiratory rate increased significantly(P<0.05), RR interval,diastolic blood pressure, mean blood pressure reduced significantly(P<0.05). Conclusion Under these study condictions,no obvious change in the cardiovascular system and the respiratory system of Beagle dogs was found after subcutaneous administration with NBZGY.

    Comparison between Water and Ethanol Extracts of Xianlinggubao for Effects on Hepatic Mitochondria

    Objective To investigate whether XLGB can result in liver injury through the change of mitochondrial function index.Methods Primary hepatocytes isolated from SD rat were treated with serial dilution (1/10~1/10000) of XLGB ethanol and water extracts for 24 h. Cellular ATP content,mitochondria membrane potential (MMP) and accumulation of fluorescent probe (TMRE) in mitochondria were used to compare and evaluate the potential liver injury induced by two extracts of XLGB.Results Only 1/10 dilution of water and ethanol extracts obviously decreased cellular ATP content and MMP in rat primary hepatocytes. Serial dilution of Ethanol extracts (1/10~1/10000) dose-dependently reduced the accumulation of TMRE in the mitochondria, while only 1/10 and 1/100 dilution of water extracts induced moderate reduction of TMRE accumulation. Conclusion In this study, ethanol extracts induced more obvious mitochondrial injury than water extracts, the latter is closer to the route of XLGB administration in human. In the serial tests of water extracts, the change of mitochondrial function was only induced by 1/10 dilution (about 50-fold clinical exposure in human), indicating that XLGB may have a wide therapeutic window in clinical use.

    Study on Correlationship between Activity of T Lymphocyte Subsets and NK Cells and Activity of Probiotics in Cancer Patients

    Objective To study the correlationship between the activity of T lymphocyte subsets and NK cells and the activity of probiotics in cancer patients. Methods The activity of T lymphocyte subsets and NK cells in the peripheral blood of cancer patients and the healthy controls were detected by using flow cytometry. The total number of bacteria and the number of lactobacillus in the feces of cancer patients and the healthy controls were detected by using plate count method. The correlationship between the activity of probiotics, T lymphocyte subsets, and NK cells and antibiotics use in cancer patients was statistically analyzed by t-test of two sample mean,one-way analysis of variance(F-test), q-test and correlation analysis, and the correlation coefficients were also examined. Results In overall comparison of cell detecting, the percentage of CD3+, CD4+, NK cells, and CD4+/CD8+ ratio in peripheral blood of cancer patients decreased significantly, while the percentage of CD8+ significantly increased, compared with the normal control group, the difference was statistically significant(each P< 0.05). In stratified comparison of cell detecting, there was no statistical difference in the percentage of CD3+, CD4+, NK cells, and CD4+/CD8+ ratio in peripheral blood between the fewer days group and the control group(each P> 0.05). Compared with the normal control group, both the moderate days group and the more days group had statistical difference in the percentage of CD3+, CD4+, CD8+, NK cells , and CD4+/CD8+ ratio in peripheral blood(each P< 0.05; 0.01). In overall comparison of bacteria detecting, The total number of bacteria of the case group increased significantly,while the number of lactobacillus significantly decreased, compared with the normal control group, the difference was statistically significant(each P< 0.050). In stratified comparison of bacteria detecting, there was no statistical difference in the total number of bacteria and the number of lactobacillus between the fewer days group and the normal control group(each P> 0.05). Compared with the normal control group,both the more days group and the moderate days group had statistical difference in the total number of bacteria and lactobacillus(P< 0.05, P<0.01). with the correlation analysis, there was a weakly positive correlation between the activity of CD3+, CD4+, CD8+, NK cells, and CD4+/CD8+ ratio in peripheral blood and the activity of lactobacillus of the cancer patients(r=0.385, 0.406, 0.371, 0.383, 0.401, each P< 0.05). Conclusion There is a weakly positive correlation between the activity of T lymphocyte subsets, NK cells and the activity of probiotics in cancer patients.

    Analysis of Endomorphin-1 and Endomorphin-2 in Peripheral Blood of Postmenpausal Women with Long-term Low-dose Hormone Replacement Therapy

    Objective To investigate the levels of EM-1 and EM-2 in peripheral blood with long-term low dose HRT therapy and analyze the relationship between endomorphins and estradiol. Methods 169 female medical staff in Peking Union Medical College Hospital were included in our study. Based on age, participants were divided into 4 groups, including 50~59 , 60~64 , 65~69 and over 70 years old , HRT group and control group were included in each age group. Plasma hormones including estradiol (E2), progesterone (P) and testosterone (T) were detected by using Bio-RAD 550 Model ELISA(enzyme-linked immunosorbent assay) device, and levels of EM-1 and EM-2 were examined by radioimmunoassay methods. Results Compared to control group at each age, plasma estrodiol (E2) levels obviously increased in HRT groups (P<0.05). The levels of EM-1 and EM-2 in HRT group of each age decreased compared to control group, meanwhile, pearson correlation analysis showed that the level of endomorphin-1 (EM-1) on all subjects was correlated with their individual estradiol(P<0.01). Conclusion This is the first observation of the relationship between endomorphins and estradiol in a long-term low-dose HRT therapy, which providing clinical significance of protecting brain cognition in postmenpausal women.

    Study on Determination of Dye Auramine O in Gegen Qinlian Tablets by HPLC-MS/MS

    Objective To establish method for determination of dye auramine O in Gegen Qinlian tablets by LC-MS/MS. Methods Agilent Eclipse plus C18(2.1×100 mm, 1.8μm) was used. A gradient elution program of acetonitrile-0.01 mol·L-1 ammonium acetate was adopted at the flow rate of 0.3 mL· min-1. The column temperature was maintained at 35℃ and the sample size was 2 μL. The analyses were detected by an electrospray ionization tandem mass spectrometry with dynamic MRM. Results The results of verification of methodology showed the calibration curve of auramine O was good linear in the range of 0.093 4~1.869 pg (r =0.999 9), and the recovery was 94.38% (RSD=3.60%, n=9). Conclusion The method was convenient, accurate with good precision, reproducibility and stability which has good linear relationship. It can be used for the determination the content of dye auramine O in Gegen Qinlian tablets.

    Objective To provide support for function adjustment of the adverse drug cluster events early warning system and improve the early warning ability. Methods More than 3 thousands early warning information in 2014 was analyzed systematically, and the risk factors influencing the early warning system were discussed. Results The risk factors included the early warning rules and algorithms, the quality of adverse drug reaction/event reports, foundation database, and the processing procedure for dealing with the early warning information of cluster adverse drug events. Conclusion The early warning system of the adverse drug cluster events can detect the product quality problem effectively. The function adjustment recommendations for the early warning system were proposed according to the identified risk factors.

    Objective To enhance emergency prevention to group adverse drug events and response ability. Methods Through introducing the emergency exercises and the specific characteristics of group adverse drug events emergency exercises, this paper discusses how to design group adverse drug events emergency exercises. Results and Conclusion Group adverse drug events emergency exercises need to be equipped with both hardwares and software systems. It is necessary to pay attention to the emergency response levels, supervision of public opinion, information management and report during the exercise design and process.

    Abstract Objective To evaluate the consistency of chemiluminescence microparticle immunoassay (CMIA) and fluorescence polarization immunoassay assay (FPIA) in determination of antiepileptic plasma drugs concentration with valproic acid as an example. Methods CMIA and FPIA methods were applied to determine the plasma concentration of valproic acid in the same sample, respectively. Then the consistency of the two methods with Passing-Bablokregression and Bland-Altman method was investigated. Results The regression equation of the two determination results by CMIA (y) and FPIA (x) was y =1.0049x-0.9851(R2=0.978,n=318), which showed the correlation of the two methods was good. Bland-Altman analysis showed that the consistency of the two methods for determining was good. Conclusion CMIA and FPIA methods for determination of valproate plasma concentrations had good correlation, but there was systematic error. In the clinical treatment, we should pay attention to make some adjustment and choice.

    Literature Analysis of 119 Cases of Adverse Drug Reactions Induced by Infliximab

    Objective To explore the general characteristics of adverse drug reactions induced by infliximab, and provide reference for safe and rational use of infliximab. Methods The articles about infliximab adverse reactions published from 2008 to 2015 in Wanfang and CNKI databases were searched, and the 119 cases documented by the articles were reviewed retrospectively. Results Primary diseases include rheumatoid arthritis, ankylosing spondylitis, ulcerative colonitis, Crohn’s disease, psoriasis, and juvenile idiopathic arthritis. Most adverse reactions occurred during the 3rd and 4th treatments. Infusion reactions were the most common. Skin, respiratory system, digestive system and coronary vascular system were often involved. Prognosis of most adverse reactions were good. Conclusion Adverse drug reactions induced by infliximab involve multiple systems and organs. Vigillance, surveillance and quick management are required.

    Objective To analyze and summarize the poisoning events induced by Datura metel L.in the literature. Methods The cases in the literature of poisoning events induced by Datura metel L. were searched and then a related database was established for statistical analysis. Results A lot of poisoning cases caused by Datura metel L. had been reported. The reasons included misuse, irrational dosage in folk prescriptions or accidentally ingested Datura metel L.by wrong prescription. Conclusion The risks of Datura metel L. should be seriously considered by related departments in order to guarantee safe drug use and safeguard the physical health of people.

    Analysis of Irrational Application of Shenmai Injections in Four Cities in Guangdong

    Objective To achieve the rational application of Shenmai injections through active monitoring and to analyze irrational application in daily practices, so as to establish the effective guidance for the basic-grade hospitals at 4 city areas of Guangdong province. Methods A method of prospective case series study was designed to inquire total of 2 402 patients who were injected Shenmai injections in 13 basic-grade hospitals at 4 city areas of Guangdong province. Results Statistic analysis indicated that the level of irrational application of Shenmai injections in the grade I hospitals 82.64% was significantly higher χ2=6.66, P=0.0099 than that in the grade II hospitals 76.45%. The major performances of irrational applications included: injections with over doses, incorrect selections of solvent, over-high concentrations and improper combinations of drugs. Over dose application rate of Shenmai injections in hospitals of the grade I and the grade II were 74.21% and 47.10%, respectively. Conclusion The irrational application of Shenmai injections in hospitals at 4 city areas of Guangdong province is a serious problem. Practical guidance in rational use of the drug and the guide for doctors in primary hospitals should be strengthened.

    Bibliometric Analysis of Erythromycin and Its Derivatives in Use of Promoting Gastric Motility

    Objective To use bibliometric analysis to do statistics and analyze domestic and international research status of erythromycin and its derivatives in use of promoting gastric motility, to expand the clinical use of erythromycin and assure rational drug use. Methods Embase,Pubmed,Web Of Science and CNKI were searched to collect all related literatures from beginning to August, 2015. Endnote X7 was used to sort out the type of literature, published year, first author, research institution, published journal, cited frequency and research contents with bibliometric methods. Results 597 literatures were included. The literatures were mainly clinical researches and reviews, others include animal experiments, case reports, retrospective studies, etc. Research content includes experimental study, clinical application, adverse effect, and precautions of erythromycin and its derivatives in use of promoting gastric motility. Conclusion Erythromycin and its derivatives in use of promoting gastric motility, which is in treatment of gastroparesis, promoting gastric emptying, especially in diabetic gastroparesis and neonatal feeding intolerance, have become the research hotspot.
